The changes of AF structure influenced vacuolar morphology. Aleurone layers were stripped from the intact seeds imbibed for 12 h, and then the isolated layers were pre-incubated in distilled water alone from 1 d to 7 d, the incubated layers were labeled with FITC-phalloidin, the organization of AF (green) was observed by LSCM, and images were immediately captured. The experiments were repeated three times, and similar images were obtained.
