Typical dot-blotting images of four representative serum specimens are shown together with immuno-dot signal intensities quantified using NIH Image-J. Seed protein extracts were spotted in duplicate on the membrane from the non-transgenic Koshihikari (NT), two lines (M1 and M2) of transgenic rice with reduced content of the major allergens (Major), and two lines (MH1 and MH2) of Major x HMW lines. Human serum albumin (H, HSA) was used as a negative antigen control. A control serum specimen with a low content of IgE to rice seed proteins was also analyzed for comparison. The dot intensities to Major and Major x HMW are shown as mean ± SD of four dots, while those of NT and HSA are mean of two dots.
