In vitro ubiquitin conjugation assays using purified proteins. After ubiquitination reactions as described, samples were subjected to SDS-PAGE and a western blot using an anti-Ub antibody was preformed to monitor poly-Ub chain formation. The background of spontaneously formed di-Ub in the absence of E2 or Uev (Lanes 1, 3, and 7) is commonly observed in these reactions. Line 2 containing GST and OsUbc13 serves as a negative control to rule out the effect of GST tag.
